Role of tyrosine 129 in the active site of spinach glycolate oxidase.
The enzymatic properties and the three-dimensional structure of spinach glycolate oxidase which has the active-site Tyr129 replaced by Phe (Y129F glycolate oxidase) has been studied. The structure of the mutant is unperturbed which facilitates interpretation of the biochemical data. Y129F glycolate oxidase has an absorbance spectrum with maxima at 364 and 450 nm (epsilon max = 11400 M-1 cm-1). ...

Spinach glycolate oxidase (GAO) was expressed in Escherichia coli using the T7 RNA polymerase promotor. The enzyme accounts for approx. 1% of the soluble protein fraction and is expressed as a soluble and active enzyme. Comparison with GAO expressed in Saccharomyces cerevisiae (Macheroux, P., Massey, V., Thiele, D.J. and Volokita, M. (1991) Biochemistry 30, 4612-4619) showed that the GAO expres...
متن کاملPreliminary crystallographic data for glycolate oxidase from spinach.
Glycolate oxidase, an enzyme that plays an important role in photorespiration in plants, has been purificant from spinach and crystallized in two different crystal forms. Form A which was obtained with tertiary butanol as precipitating agent belongs to space group I 422 with unit cell dimensions a = b = 148.1 A and c = 134.9 A. This form diffracts to high resolution and will be used for further...
متن کاملThe evolution of photorespiratory glycolate oxidase activity.
The first step of photosynthetic carbon fixation is catalyzed by ribulose-1,5bisphosphate carboxylase/oxygenase, which arose when the atmosphere contained virtually no O2. The increase of atmospheric O2 caused by oxygenic photosynthesis gave rise to photorespiratory metabolism, a process whereby O2 substitutes for CO2, causing ribulose-1,5bisphosphate carboxylase/oxygenase to produce the toxic ...

The g1lycolate oxidase reactioni by x-hich gly colate is oxidized to glyoxylate (3, 10) is catailyzed by a flavoprotein (18) th,at is widely distributed in higher plants (12) and fungi (4). Its presence is g,elleralllly assumed in all phoiosynithetic tissuies, since glycolate is an early product of photosynthesis (14), yet this substrate is tusually found in oiily minute amounits. The oxidation...
